FAM
• High quantum yields. Better stability than fluorescein in various buffer conditions.
• There are two isomers of FAM (carboxyfluorescein): 5-FAM and 6-FAM.
• FAM can be introduced as phosphoramidite (mixture of 5/6-FAM isomers), and post-synthetically as mixed, 5- or 6-FAM isomer.
• 3’-(6-FAM) allows effective blockage of the 3’-terminus from polymerase extension as well as exonuclease digestion.
• These oligos are tested by U.V. absorbance monitoring, and calculation of A260/A494 ratio.
