Analysis of human gene expression levels
With the completion of the Humane Genome Project, the functions of genes need to be determined, so studies of gene expression become the focus.
The introduction of Real-Time PCR has made it possible to accurately quantify starting amounts of nucleic acids during a PCR reaction without the need for post-PCR analyses. Measurements of PCR products can be taken during the linear phase of the amplification reactions, which allows an accurate quantification of the template(s) of interest.
Housekeeping genes are often used as controls in the quantitative analysis of gene expression. By using a control as a reference, one can normalize for differences in the amount of total RNA added to each reaction.
