Achieve longer qPCR probe designs with reduced background fluorescence
Designing probes with a high-enough Tm on AT rich regions often result in longer sequences. Traditional Dual Labelled probes with sequences longer than 30 bases will have less efficient quenching and can be non-functional or exhibit increased background fluorescence signal within the qPCR reaction.
Introducing a second internal quencher in the probe will reduce the background fluorescence and often improves overall qPCR reaction sensitivity and consistency.
